Secondary antibodies
Secondary antibodies are antibodies that bind to the protein of interest on a primary antibody in the indirect detection process. Secondaries are usually conjugated to fluorophores, enzymes (eg HRP, alkaline phosphatase), or proteins (eg biotin). They are produced by immunizing a host animal with antibodies from a different species. For example, immunizing a goat with rabbit IgG would generate goat anti-rabbit IgG secondary antibodies.
When selecting a secondary antibody for your experiment, it should match the species the primary antibody was raised in and the isotype of the primary antibody of interest. For instance, the goat anti-rabbit IgG secondary antibody mentioned above can be used to detect the signal of a rabbit IgG primary antibody.
